What is Ozette Resolve?
Ozette’s Resolve is a cloud-native solution that adaptively unmixes spectral cytometry data by adapting to variable autofluorescence and fluorescent spectra across cells as well as estimating background and transformation parameters for each fluor in the panel. This eliminates negative fluorescence and optimizes the unmixed measurements, which can improve marker resolution and eliminates the need for manual compensation.
Key Concepts
- Instrument: The specific instrument and instrument configuration used to collect data.
- Panels: The highest level of organization – defined by instrument configuration, fluorophores and materials types
- Control Files: Single-color controls, unstained controls and full stained assay controls used by the unmixing algorithm
- Control Sets: Curated collections of control files used to unmix multicolor data
- Gating: Manual selection of cell populations to optimize unmixing performance
- Event-Level Spectral Signature Plot: Visualization available within the gating page that displays individual spectral signatures for each event
- Unmixing: The computational process that transforms event measurements from measurements taken on instrument detectors to measurements corresponding to markers in a panel
